Immunohistochemistry (IHC)

(Immunohistochemistry analysis of Phospho-IRS1 [pY612] showing staining in the cytoplasm and nucleus of paraffin-embedded human skeletal muscle tissue (right) compared to a negative control without primary antibody (left). To expose target proteins, antigen retrieval was performed using 10mM sodium citrate (pH 6.0), microwaved for 8-15 min. Following antigen retrieval, tissues were blocked in 3% H2O2-methanol for 15 min at RT, washed with ddH2O and PBS, and then probed with MBS624304 diluted in 3% BSA-PBS at a dilution of 1:20 overnight at 4°C in a humidified chamber. Tissues were washed extensively in PBST and detection was performed using an HRP-conjugated secondary antibody followed by colorimetric detection using a DAB kit. Tissues were counterstained with hematoxylin and dehydrated with ethanol and xylene to prep for mounting.)

Flow Cytometry (FC/FACS)

(Flow cytometry analysis of IRS1 [pY612] was done on MCF7 cells treated with Insulin (100nM, 10 minutes). Cells were fixed with 70% ethanol for 10 minutes, permeabilized with 0.25% Triton™ X-100 for 20 minutes, and blocked with 5% BSA for 30 minutes at RT. Cells were labeled with MBS624304 (red histogram) or with rabbit isotype control (pink histogram) at 3-5ug/million cells in 2.5% BSA. After incubation at RT for 2 hours, the cells were labeled with Alexa Fluor® 488 Goat Anti-Rabbit Secondary Antibody at a dilution of 1:400 for 30 minutes at RT. The purple histogram represents unstained control cells and the green histogram represents no-primary-antibody control.)

Related Product Information for anti-IRS1 antibody
Insulin receptor substrates (IRS) are responsible for several insulin related activities, such as glucose homeostasis, cell growth, cell transformation, apoptosis and insulin signal transduction. Serine/threonine phosphorylation of IRS-1 has been demonstrated to be a negative regulator of insulin signaling and is responsible for its degradation, although IRS-1 degradation pathways are not well understood. IRS-1 has also been shown to be constitutively activated in cancers such as breast cancer, Wilm"e;s tumors, and adrenal cortical carcinomas, thus making IRS-1 phosphorylation and subsequent degradation an attractive therapeutic option. To date there have been four subtypes identified: IRS-1,2,3, and 4, with IRS-1 being widely expressed.

NCBI Description

This gene encodes a protein which is phosphorylated by insulin receptor tyrosine kinase. Mutations in this gene are associated with type II diabetes and susceptibility to insulin resistance. [provided by RefSeq]

Uniprot Description

IRS1: an adaptor protein that is one of the major substrates of the insulin receptor kinase. Contains multiple tyrosine phosphorylation motifs that serve as docking sites for SH2-domain-containing proteins including phosphatidylinositol 3-kinase p85 subunit and GRB-2. Phosphorylated on ser/thr residues by kinases including JNK and IKK.

Product Notes

The IRS1 irs1 (Catalog #AAA624304) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The IRS-1, phosphorylated (Tyr612) (Insulin Receptor Substrate 1) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's IRS-1 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC), Western Blot (WB), Immunohistochemistry (IHC). Other applications not tested. Recommended Dilution: Immunohistochemistry (Paraffin): 1:10-1:100 Flow Cytometry: 3-5ug/million cells Optimal dilutions to be determined by the researcher. Researchers should empirically determine the suitability of the IRS1 irs1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IRS-1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
