On the basis of both functional and structural considerations, members of the IkappaB family of proteins can be divided into four groups. The first of these groups, IkappaB-alpha, includes the avian protein pp40 and the mammalian MAD-3, both of which inhibit binding of p50-p65 NFkB complex or Rel protein to their cognate binding sites but do not inhibit the binding of p50 homodimer to kappaB sites, suggesting that the IkappaB-alpha family binds to the p65 subunit of p50-p65 heterocomplex through ankyrin repeats. The second member of the IkappaB family is represented by a protein designated IkappaB-beta. The third group of IkappaB proteins is represented by IkappaB-gamma, which is identical in sequence with the C-terminal domain of the p110 precursor of NFkB p50 and is expressed predominantly in lymphoid cells. An additional IkappaB family member, IkappaB-epsilon, has several phosphorylated forms and is primarily found complexed with Rel A and/or c-Rel.

NCBI Description

The protein encoded by this gene binds to components of NF-kappa-B, trapping the complex in the cytoplasm and preventing it from activating genes in the nucleus. Phosphorylation of the encoded protein targets it for destruction by the ubiquitin pathway, which activates NF-kappa-B by making it available to translocate to the nucleus. IkB-epsilon: Inhibits NF-kappa-B by complexing with and trapping it in the cytoplasm. Inhibits DNA-binding of NF-kappa-B p50-p65 and p50-c-Rel complexes. Interacts with RELA, REL, NFKB1 nuclear factor NF-kappa-B p50 subunit and NFKB2 nuclear factor NF-kappa-B p52 subunit. Highly expressed in spleen, testis and lung, followed by kidney, pancreas, heart, placenta and brain. Also expressed in granulocytes and macrophages. Belongs to the NF-kappa-B inhibitor family.
