This item recognises the glutamate receptor 1 (also known as AMPA) which acts as an excitatory neurotransmitter at many synapses in the central nervous system. Binding of the excitatory neurotransmitter L-glutamate induces a conformation change, leading to the opening of the cation channel, and thereby converts the chemical signal to an electrical impulse. The receptor then desensitizes rapidly and enters a transient inactive state, characterized by the presence of bound agonist.This antibody linked to immobilized Protein A immunoprecipitates detergent-solubilized receptor from brain or transfected cells.

Application Notes
Western Blot: Western blot analysis of rat brain shows a single band of approximately 110 kDa co-migrating with GluR1 expressed in transfected cells.
Immunohistology: Paraformaldehyde or paraformaldehyde/glutaraldehyde are suitable. Cryostat and vibratome sections can be used with or without Triton X-100 treatment.
Immunohistology - Frozen: Minimum Dilution: 1 ug/ml; Maximum Dilution: 3 ug/ml
Western Blotting: Minimum Dilution: 0.5 ug/ml; Maximum Dilution: 2.5 ug/ml

GluR1: an integral membrane protein belonging to the glutamate-gated ion channel family. L-glutamate (Glu) acts as an excitatory neurotransmitter at many synapses in the central nervous system. Glutamate receptors are heteromeric protein complexes with multiple subunits, each possessing transmembrane regions, and all arranged to form a ligand-gated ion channel. The postsynaptic actions of Glu are mediated by a variety of receptors that are named according to their selective agonists. Each of the four GluR proteins (GRIA1-4) include flip and flop isoforms generated by alternative RNA splicing. Expressed in granule and pyramidal cells in the hippocampal formation.
