WB: 1:500-1:2,000 dilution
Note: Many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt/Sonication Protocol when preparing nuclear extracts for Western blot.
Background: GLI1 (Glioma-associated oncogene 1) is a zinc finger transcription factor and oncogene. GLI1 is part of the hedgehog signaling pathway, but is also activated in response to TGF-b signaling as well. GLI1 activates transcription of a number of genes that promote cell cycle progression. GLI1 is overexpressed as a result of constitutive activation of hedgehog signaling, which plays a role in the initiation of cancer. GLI1 also promotes self-renewal of neural stem cells.
NCBI and Uniprot Product Information
Uniprot Description
GLI1: Acts as a transcriptional activator. May regulate the transcription of specific genes during normal development. May play a role in craniofacial development and digital development, as well as development of the central nervous system and gastrointestinal tract. Mediates SHH signaling and thus cell proliferation and differentiation. Interacts with KIF7. Interacts with ZIC1; the interaction enhances transcription activation. Amplified in glioblastoma cells. Testis, myometrium and fallopian tube. Also expressed in the brain with highest expression in the cerebellum, optic nerve and olfactory tract. Belongs to the GLI C2H2-type zinc-finger protein family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes
The GLI1 gli1 (Catalog #AAA388812) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The GLI1 antibody (pAb) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's GLI1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Applications Validated: WB: 1:500-1:2,000 dilution Note: Many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt/Sonication Protocol when preparing nuclear extracts for Western blot. Researchers should empirically determine the suitability of the GLI1 gli1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "GLI1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
