Application Notes
Peptide ELISA: Antibody detection limit dilution 1: 8000.
Immunohistochemistry: In paraffin embedded Human Thyroid Gland shows nuclear staining in thyrocytes. Recommended concentration, 2-4ug/ml.
Western Blot: Preliminary experiments gave an approx 70kDa band in Human Thymus and Thyroid Gland lysates after 0.3ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 38.1kDa according to NP_004464.2. The 70kDa band was successfully blocked by incubation with the immunizing peptide.

NCBI Description

This intronless gene belongs to the forkhead family of transcription factors, which is characterized by a distinct forkhead domain. This gene functions as a thyroid transcription factor which likely plays a crucial role in thyroid morphogenesis. Mutations in this gene are associated with congenital hypothyroidism and cleft palate with thyroid dysgenesis. The map localization of this gene suggests it may also be a candidate gene for squamous cell epithelioma and hereditary sensory neuropathy type I. [provided by RefSeq, Jul 2008]

Uniprot Description

FOXE1: Probable transcription factor. Could be involved in thyroid gland organogenesis. Defects in FOXE1 are the cause of Bamforth-Lazarus syndrome (BLS). BLS is associated with thyroid agenesis, cleft palate and choanal atresia.

Protein type: DNA-binding

Chromosomal Location of Human Ortholog: 9q22

Cellular Component: nucleus

Molecular Function: DNA binding; sequence-specific DNA binding; transcription factor activity

Biological Process: anatomical structure morphogenesis; cell migration; embryonic organ morphogenesis; hair follicle morphogenesis; hard palate development; negative regulation of transcription from RNA polymerase II promoter; negative regulation of transcription, DNA-dependent; positive regulation of transcription, DNA-dependent; soft palate development; thymus development; thyroid gland development; thyroid hormone generation; transcription, DNA-dependent

The Goat anti-FOXE1/TTF2 Antibody reacts with Tested: Human; Expected from sequence similarity: Human and may cross-react with other species as described in the data sheet. AAA Biotech's FOXE1/TTF2 can be used in a range of immunoassay formats including, but not limited to, Peptide ELISA (EIA), Immunohistochemistry (IHC). Peptide ELISA: Antibody detection limit dilution 1: 8000. Immunohistochemistry: In paraffin embedded Human Thyroid Gland shows nuclear staining in thyrocytes. Recommended concentration, 2-4ug/ml. The 70kDa band was successfully blocked by incubation with the immunizing peptide. Researchers should empirically determine the suitability of the FOXE1 foxe1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: AYPGGIDRFV SAM. It is sometimes possible for the material contained within the vial of "FOXE1/TTF2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
