Rabbit DNA Polymerase theta Polyclonal Antibody | anti-POLQ antibody
DNA Polymerase theta Antibody
IF/ICC: 1:100-1:500,
IHC: 1:50-1:200,
ELISA(peptide): 1:20000-1:40000
*The optimal dilutions should be determined by the end user.
IMPORTANT: For western blots, incubate membrane with diluted antibody in 5% w/v milk , 1X TBS, 0.1% Tween®20 at 4°C with gentle shaking, overnight.
Immunofluorescence/Immunocytochemistry (ICC/IF)
(MBS9612322 staining HepG2 cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab (MBS9612322) and mouse anti-beta tubulin Ab(MBS9610328) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Western Blot (WB)
(Western blot analysis of extracts from Mouse spleen, using DNA Polymerase theta Antibody. The lane on the left was treated with blocking peptide.)
Immunohistochemistry (IHC)-Paraffin
(MBS9612322 at 1/100 staining Rat testis tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary antibody at 4°C overnight. An HRP conjugated anti-Rabbit antibody was used as the secondary antibody.)
NCBI and Uniprot Product Information
Uniprot Description
POLQ: Has a DNA polymerase activity on nicked double-stranded DNA and on a singly primed DNA template. The enzyme activity is resistant to aphidicolin, and inhibited by dideoxynucleotides. Exhibites a single-stranded DNA-dependent ATPase activity. Could be involved in the repair of interstrand cross-links. Belongs to the DNA polymerase type-A family. 2 isoforms of the human protein are produced by alternative splicing.
Protein type: DNA repair, damage; Transferase; EC 2.7.7.7
Chromosomal Location of Human Ortholog: 3q13.33
Cellular Component: nucleoplasm
Molecular Function: DNA-directed DNA polymerase activity; damaged DNA binding; ATP binding; single-stranded DNA-dependent ATPase activity
Biological Process: DNA repair; DNA-dependent DNA replication

AAA Biotech's DNA Polymerase theta can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Western Blot (WB), Immunofluorescence (IF), Immunocytochemistry (ICC), Immunohistochemistry (IHC). WB: 1:500-1:2000, IF/ICC: 1:100-1:500, IHC: 1:50-1:200, ELISA(peptide): 1:20000-1:40000 *The optimal dilutions should be determined by the end user. IMPORTANT: For western blots, incubate membrane with diluted antibody in 5% w/v milk, 1X TBS, 0.1% Tween®20 at 4°C with gentle shaking, overnight. . Researchers should empirically determine the suitability of the POLQ polq for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DNA Polymerase theta, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
