This antibody was developed against a mixture of KLH-conjugated synthetic peptides corresponding to amino acids 25-44 and 113-130 of human DUT-N.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Deoxyuridine triphosphate nucleotidohydrolase (dUTPase) is a ubiquitous enzyme that functions in the hydrolysis of dUTP to dUMP and pyrophosphate. Canman and co-workers have demonstrated that, in certain human tumor cell lines, increased levels of dUTPase are responsible for an increase in resistance to the cancer chemotherapeutic agent fluorodeoxyuridine (FUdR), a thymidine synthase inhibitor. Two distinct forms of dUTPase exist in humans. Cellular fractionation experiments suggest that the more abundant, lower mass form of dUTPase (DUT-N) localizes in the nucleus, while the higher mass form (DUT-M) is associated with the mitochondria.

NCBI Description

This gene encodes an essential enzyme of nucleotide metabolism. The encoded protein forms a ubiquitous, homotetrameric enzyme that hydrolyzes dUTP to dUMP and pyrophosphate. This reaction serves two cellular purposes: providing a precursor (dUMP) for the synthesis of thymine nucleotides needed for DNA replication, and limiting intracellular pools of dUTP. Elevated levels of dUTP lead to increased incorporation of uracil into DNA, which induces extensive excision repair mediated by uracil glycosylase. This repair process, resulting in the removal and reincorporation of dUTP, is self-defeating and leads to DNA fragmentation and cell death. Alternative splicing of this gene leads to different isoforms that localize to either the mitochondrion or nucleus. A related pseudogene is located on chromosome 19. [provided by RefSeq]
