NCBI Description

DAD1, the defender against apoptotic cell death, was initially identified as a negative regulator of programmed cell death in the temperature sensitive tsBN7 cell line. The DAD1 protein disappeared in temperature-sensitive cells following a shift to the nonpermissive temperature, suggesting that loss of the DAD1 protein triggered apoptosis. DAD1 is believed to be a tightly associated subunit of oligosaccharyltransferase both in the intact membrane and in the purified enzyme, thus reflecting the essential nature of N-linked glycosylation in eukaryotes. [provided by RefSeq, Jul 2008]

Uniprot Description

DAD1: Component of the N-oligosaccharyl transferase enzyme which catalyzes the transfer of a high mannose oligosaccharide from a lipid-linked oligosaccharide donor to an asparagine residue within an Asn-X-Ser/Thr consensus motif in nascent polypeptide chains. N-glycosylation occurs cotranslationally and the complex associates with the Sec61 complex at the channel-forming translocon complex that mediates protein translocation across the endoplasmic reticulum (ER). Loss of the DAD1 protein triggers apoptosis. Belongs to the DAD/OST2 family.

Protein type: Endoplasmic reticulum; Transferase; Glycan Metabolism - N-glycan biosynthesis; Membrane protein, integral; Apoptosis; Membrane protein, multi-pass; EC 2.4.99.18

Chromosomal Location of Human Ortholog: 14q11.2

Cellular Component: endoplasmic reticulum membrane; membrane; oligosaccharyl transferase complex; integral to membrane

Molecular Function: dolichyl-diphosphooligosaccharide-protein glycotransferase activity; oligosaccharyl transferase activity

Biological Process: response to drug; blastocyst development; cellular protein metabolic process; apoptosis; protein amino acid glycosylation; protein amino acid N-linked glycosylation via asparagine; post-translational protein modification; response to nutrient; negative regulation of apoptosis

Product Notes

The DAD1 dad1 (Catalog #AAA6472164) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Defender of Apoptotic Cell Death 1 (DAD1) (APC) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Defender of Apoptotic Cell Death 1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). WB: 0.5-2ug/ml, detects a band of approximately 22kD in HepG2 cell lysates. Applications are based on unconjugated antibody. Researchers should empirically determine the suitability of the DAD1 dad1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Defender of Apoptotic Cell Death 1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
