CUEDC2 is a 287aa protein belonging to the CUEDC2 family with a CUE domain. CUEDC2 is known to interact with PGR and ESR1 with the help of its CUE domain. The exact function of CUEDC2 is yet unknown, but reports suggest that it interacts with PR and promotes progesterone-induced PR degradation by the ubiquitin-proteasome pathway. CUEDC2 participates in ubiquitination and subsequent proteasomal degradation of PGR and ESR1 and thus control their protein levels and plays an important role in breast cancer proliferation. Studies on CUEDC2 suggest that it acts as an adaptor protein that interacts with IKK and regulates IKK dephosphorylation by recruiting PP1.

Uniprot Description

Function: Down-regulates ESR1 protein levels through the ubiquitination-proteasome pathway, regardless of the presence of 17 beta-estradiol. Also involved in 17 beta-estradiol-induced ESR1 degradation. Controls PGR protein levels through a similar mechanism. Ref.6 Ref.9

Subunit structure: Interacts with PGR. Interacts with ESR1 in the presence or absence of 17 beta-estradiol. Ref.6 Ref.9

Subcellular location: Cytoplasm. Nucleus Ref.6.

Tissue specificity: Significantly up-regulated in breast tumor tissues compared with matched adjacent normal tissues (at protein level). Levels inversely correlate with ESR1 in breast cancers and are lower in low-grade tumors compared to high-grade tumors.

Domain: The CUE domain mediates interaction with PGR and ESR1.

Involvement in disease: May predict the clinical outcome of tamoxifen therapy of breast cancer patients. Patients with tumors that highly express CUEDC2 do not respond to tamoxifen treatment as effectively as those with tumors with low expression.
