Related Product Information for anti-GJA5 antibody
Connexin 40 (Cx40) is a gap junction protein highly expressed by vascular endothelial cells and in the mammalian heart, specifically in the atrial myocardium and the His-Purkinje conduction system. At least three other connexins are expressed in the heart, including Cx37 (endocardium), Cx43 (atrial and ventricular myocardium, distal parts of the conduction system), and Cx45 (throughout the heart). Mice lacking the gene for Cx40 exhibit reduced atrial but normal ventricular conduction velocity; these mice display right bundle-branch block and impaired left bundle-branch conduction. Cx40 and Cx43 frequently co-localize in cells of the cardiovascular system, and participate in the formation of heterotypic gap junction channels. For example, atrial myocytes express both Cx40 and Cx43, which oligomerize into the same hemichannel. Atrial connexons consist of hexomeric connexin assemblies that contain varying proportions of Cx40 and Cx43 proteins.

NCBI Description

This gene is a member of the connexin gene family. The encoded protein is a component of gap junctions, which are composed of arrays of intercellular channels that provide a route for the diffusion of low molecular weight materials from cell to cell. Mutations in this gene may be associated with atrial fibrillation. Alternatively spliced transcript variants encoding the same isoform have been described. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: One gap junction consists of a cluster of closely packed pairs of transmembrane channels, the connexons, through which materials of low MW diffuse from one cell to a neighboring cell.

Subunit structure: A connexon is composed of a hexamer of connexins.

Subcellular location: Cell membrane; Multi-pass membrane protein. Cell junction › gap junction.

Involvement in disease: Familial atrial standstill (FAS) [MIM:108770]: Atrial standstill is an extremely rare arrhythmia, characterized by the absence of electrical and mechanical activity in the atria. Electrocardiographically, it is characterized by bradycardia, the absence of P waves, and a junctional narrow complex escape rhythm.Note: The disease may be caused by mutations affecting distinct genetic loci, including the gene represented in this entry. A rare GJA5 genotype has been detected in combination with a mutation in SCN5A in a large family with atrial standstill. Ref.8Familial atrial fibrillation 11 (ATFB11) [MIM:614049]: A familial form of atrial fibrillation, a common sustained cardiac rhythm disturbance. Atrial fibrillation is characterized by disorganized atrial electrical activity and ineffective atrial contraction promoting blood stasis in the atria and reduces ventricular filling. It can result in palpitations, syncope, thromboembolic stroke, and congestive heart failure.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.9

Sequence similarities: Belongs to the connexin family. Alpha-type (group II) subfamily.

Product Notes

The GJA5 gja5 (Catalog #AAA646454) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Connexin 40, CT (CX40, Gap Junction Alpha 5 Protein, CXA-5) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Connexin 40 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunoprecipitation (IP), Immunohistochemistry (IHC), Immunofluorescence (IF). Suitable for use in Western Blot, Immunofluorescence, Immunoprecipitation and Immunohistochemistry. Dilution: Western Blot: 1-3ug/ml Immunofluorescence: 1-3ug/ml Immunoprecipitation: 7ug/reaction Immunohistochemistry (Frozen): 1-3ug/ml; Mice were perfused transcardially with 3ml pre-fixative (50mM phosphate buffer, 0.1% sodium nitrite, 1 unit/ml heparin), followed by 40ml fixative (0.16M sodium phosphate buffer, 0.2% picric acid, 1-, 2-, or 4% paraformaldehyde) and perfusion with 10ml sucrose wash (10% sucrose in 25mM sodium phosphate buffer, pH 7.4). Tissues were then stored in cryoprotectant (10% sucrose, 25mM phosphate buffer, pH 7.4, 0.04% sodium azide) for a minimum of 24 hours prior to sectioning. Researchers should empirically determine the suitability of the GJA5 gja5 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Connexin 40, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
