Western Blot (WB) (Figure 1. Western blot analysis of Collagen II using anti-Collagen II antibody. Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: rat liver tissue lysates,Lane 2: mouse liver tissue lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Collagen II antigen affinity purified polyclonal antibody at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for Collagen II at approximately 142KD. The expected band size for Collagen II is at 142KD.)

Related Product Information for anti-COL2A1 antibody
Description: Rabbit IgG polyclonal antibody for Collagen II detection. Tested with WB in Human; Mouse; Rat.
Background: Collagen, type II, alpha 1, also known as COL2A1, is a human gene which is the major collagen synthesized by chondrocvtes. COL2A1 is mapped to 12q13.11. This gene encodes the alpha-1 chain of type II collagen, a fibrillar collagen found in cartilage and the vitreous humor of the eye. Mutations in this gene are associated with achondrogenesis, chondrodysplasia, early onset familial osteoarthritis, SED congenita, Langer-Saldino achondrogenesis, Kniest dysplasia, Stickler syndrome type I, and spondyloepimetaphyseal dysplasia Strudwick type. In addition, defects in processing chondrocalcin, a calcium binding protein that is the C-propeptide of this collagen molecule, are also associated with chondrodysplasia.

NCBI Description

There are two transcripts identified for this gene. [provided by RefSeq, Jul 2008]

Uniprot Description

Type II collagen is specific for cartilaginous tissues. It is essential for the normal embryonic development of the skeleton, for linear growth and for the ability of cartilage to resist compressive forces.

Product Notes

The COL2A1 col2a1 (Catalog #AAA1751255) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Collagen II/COL2A1 Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Collagen II/COL2A1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). WB: 0.1-0.5 mug/ml Tested Species: In-house tested species with positive results. Researchers should empirically determine the suitability of the COL2A1 col2a1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Collagen II/COL2A1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
