Western Blot (WB)
(Western blot analysis of CMA1 using anti- CMA1 antibody (MBS176889).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: human PC-3 whole cell lysates,Lane 2: human HepG2 whole cell lysates,Lane 3: rat liver tissue lysates,Lane 4: mouse HEPA1-6 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti- CMA1 antigen affinity purified polyclonal antibody (Catalog # MBS176889) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # MBS176460) with Tanon 5200 system. A specific band was detected for CMA1 at approximately 35KD. The expected band size for CMA1 is at 27KD.)
Background: Chymase is a major secreted protease of mast cells with suspected roles in vasoactive peptide generation, extracellular matrix degradation, and regulation of gland secretion. This gene product is a chymotryptic serine proteinase that belongs to the peptidase family S1. Chymase is mapped to 14q12. It is expressed in mast cells and thought to function in the degradation of the extracellular matrix, the regulation of submucosal gland secretion, and the generation of vasoactive peptides. In the heart and blood vessels, this protein, rather than angiotensin converting enzyme, is largely responsible for converting angiotensin I to the vasoactive peptide angiotensin II.
NCBI and Uniprot Product Information
NCBI Description
This gene product is a chymotryptic serine proteinase that belongs to the peptidase family S1. Angiotensin II has been implicated in blood pressure control and in the pathogenesis of hypertension, cardiac hypertrophy, and heart failure. Thus, this gene product is a target for cardiovascular disease therapies. This gene maps to 14q11.2 in a cluster of genes encoding other proteases. [provided by RefSeq, Jul 2008]

Product Notes
The CMA1 cma1 (Catalog #AAA176889) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-CMA1 Antibody reacts with Reacts with Human. No cross reactivity with other proteins and may cross-react with other species as described in the data sheet. AAA Biotech's CMA1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC) Paraffin. Western Blot: Concentration: 0.1-0.5 ug/ml Tested Species: Human IHC (Paraffin-embedded Section): Concentration: 0.5-1 ug/ml Tested Species: Human Antigen Retrieval: By Heat WB: The detection limit for CMA1 is approximately 0.25ng/lane under reducing conditions. Optimal dilutions should be determined by the end users. Researchers should empirically determine the suitability of the CMA1 cma1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CMA1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
