Western Blot (WB)
(Western blot analysis of lysate of human CD14+ peripheral blood mononuclear cells (PBMC). PVDF Membrane was probed with 1ug/ml of MBS647676 followed by HRP-conjugated sheep IgG secondary antibody. A specific band was detected for CLEC9a at approximately 45kD. This experiment was conducted under reducing conditions. The following buffers were used: blotting buffer: 25mM Tris, pH 7.4, 0.5M NaCl and 0.05% Tween-20; blocking solution: 3% BSA in blotting buffer, pH adjusted to 7.4; antibody solution: 2% dry non-fat dry milk in blotting buffer, pH adjusted to 7.4.)

(IHC staining of immersion fixed, paraffin-embedded sections of human spleen using MBS647676 at 15ug/ml overnight at 4°C. Tissue was stained using the anti-sheep HRP-DAB Cell & Tissue Staining Kit and counterstained with hematoxylin (blue). Specific staining was localized to the cytoplasm.)
