Related Product Information for anti-CLCN3 antibody
Chloride is a critical component of all living cells. Voltage-gated chloride channels regulate cellular traffic of chloride ion. The chloride channels (CIC or CLC) performs several functions including the regulation of cell volume, membrane potential stabilization, signal transduction, and transepithelial transport. Mutations in CIC genes have been linked with several human diseases including myotonias (Thomsen's disease), cystic fibrosis, Bartters syndrome type III, Dent's disease, and X-linked recessive nephrolithiasis. In mammals, CLC proteins form a superfamily of at least 9 different genes (CLC1-7 also known as CLCN1-7 and CLK1-2 or CLCKa and CLCKb). Additional forms of these proteins are obtained by alternative splicing. All CLC proteins (~700-1000aa) are predicted to contain 10 (possibly 12) transmembrane domains. Except CLC-1 and CLC-K1/K2 that are specific for kidney, most other CLC are widely distributed in various tissues. Rat CLC-3 is 760aa membrane protein (human CLC-3 760 or 820aa; mouse CLC-3 760/818aa) (1). Alternatively spliced short forms of mouse and human CLC-3 have also been found. CLC-3 is abundant in brain (olfactory, hippocampus, and cerebellum) and moderate levels are found in the lung, kidney, and adrenals. CLC-3 has been implicated in neuronal cell function through regulation of membrane excitability by protein kinase C. It could also help neuronal cells to establish short-term memory.

NCBI Description

This gene encodes a member of the voltage-gated chloride channel (ClC) family. The encoded protein is present in all cell types and localized in plasma membranes and in intracellular vesicles. It is a multi-pass membrane protein which contains a ClC domain and two additional C-terminal CBS (cystathionine beta-synthase) domains. The ClC domain catalyzes the selective flow of Cl- ions across cell membranes, and the CBS domain may have a regulatory function. This protein plays a role in both acidification and transmitter loading of GABAergic synaptic vesicles, and in smooth muscle cell activation and neointima formation. This protein is required for lysophosphatidic acid (LPA)-activated Cl- current activity and fibroblast-to-myofibroblast differentiation. The protein activity is regulated by Ca(2+)/calmodulin-dependent protein kinase II (CaMKII) in glioma cells. Multiple alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Aug 2011]

Uniprot Description

CLCN3: Mediates the exchange of chloride ions against protons. Functions as antiporter and contributes to the acidification of the endosome and synaptic vesicle lumen, and may thereby affect vesicle trafficking and exocytosis. May play an important role in neuronal cell function through regulation of membrane excitability by protein kinase C. It could help neuronal cells to establish short-term memory. Belongs to the chloride channel (TC 2.A.49) family. ClC-3/CLCN3 subfamily. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The CLCN3 clcn3 (Catalog #AAA615873) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The CLC3 (Clcn3, Chloride Channel) reacts with Rat and may cross-react with other species as described in the data sheet. AAA Biotech's CLC3 can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB). Suitable for use in ELISA and Western Blot. Dilution: Western Blot: 1-10ug/ml (ECL). ELISA: 0.5-1ug/ml. Control peptide can be used to coat ELISA plates at 1ug/ml. Researchers should empirically determine the suitability of the CLCN3 clcn3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CLC3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
