Western Blot (WB) (Figure 1. Western blot analysis of CHRNA4 using anti-CHRNA4 antibody (MBS1750756). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: human Hela cell lysate,Lane 2: human COLO-320 cell lysate,Lane 3: human HepG2 cell lysate,Lane 4: human PANC-1 cell lysate,Lane 5: human 22RV1 cell lysate,Lane 6: human SGC-7901 cell lysate,Lane 7: rat spleen tissue lysate,Lane 8: mouse spleen tissue lysate. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CHRNA4 antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CHRNA4 at approximately 70KD. The expected band size for CHRNA4 is at 70KD. )

NCBI Description

This gene encodes a nicotinic acetylcholine receptor, which belongs to a superfamily of ligand-gated ion channels that play a role in fast signal transmission at synapses. These pentameric receptors can bind acetylcholine, which causes an extensive change in conformation that leads to the opening of an ion-conducting channel across the plasma membrane. This protein is an integral membrane receptor subunit that can interact with either nAChR beta-2 or nAChR beta-4 to form a functional receptor. Mutations in this gene cause nocturnal frontal lobe epilepsy type 1. Polymorphisms in this gene that provide protection against nicotine addiction have been described. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Feb 2012]

Uniprot Description

After binding acetylcholine, the AChR responds by an extensive change in conformation that affects all subunits and leads to opening of an ion-conducting channel across the plasma membrane permeable to sodium ions.

Product Notes

The CHRNA4 chrna4 (Catalog #AAA1750756) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-CHRNA4 Picoband antibody reacts with Human, Mouse, Rat No cross reactivity with other proteins. and may cross-react with other species as described in the data sheet. AAA Biotech's CHRNA4 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). WB: 0.1-0.5mug/ml. Researchers should empirically determine the suitability of the CHRNA4 chrna4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CHRNA4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
