Rabbit CHCHD4 Polyclonal Antibody | anti-CHCHD4 antibody
CHCHD4 Antibody
Phosphate buffered saline, pH 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol.
IHC: 1:50-1:200
Immunohistochemistry (IHC)
(MBS9603592 at 1/100 staining Human lymph cancer tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the antibody for 1.5 hours at 22 degree C. An HRP conjugated goat anti-rabbit antibody was used as the secondary.)
Subunit Structure: Monomer (PubMed:19182799). Can form homooligomers (PubMed:16185709). Interacts with GFER and forms transient disulfide bonds with GFER (PubMed:23186364, PubMed:23676665). Interacts with MICU1 (PubMed:26387864). Interacts with COX19 forming transient intermolecular disulfide bridges (PubMed:23676665).
Post-translational Modifications: Forms intrachain disulfide bridges, but exists in different redox states.
Similarity: The CHCH domain contains a conserved twin Cys-X9-Cys motif which is required for import and stability of MIA40 in mitochondria.
NCBI and Uniprot Product Information
Predicted: 16 kDa
NCBI Description
CHCHD4, a component of human mitochondria, belongs to a protein family whose members share 6 highly conserved cysteine residues constituting a -CXC-CX(9)C-CX(9)C- motif in the C terminus (Hofmann et al., 2005 [PubMed 16185709]).[supplied by OMIM, Mar 2008]
Uniprot Description
Functions as chaperone and catalyzes the formation of disulfide bonds in substrate proteins, such as COX17, COX19 and MICU1 (PubMed:16185709, PubMed:26387864, PubMed:19182799, PubMed:21059946, PubMed:23186364, PubMed:23676665). Required for the import and folding of small cysteine-containing proteins (small Tim) in the mitochondrial intermembrane space (IMS). Precursor proteins to be imported into the IMS are translocated in their reduced form into the mitochondria. The oxidized form of CHCHD4/MIA40 forms a transient intermolecular disulfide bridge with the reduced precursor protein, resulting in oxidation of the precursor protein that now contains an intramolecular disulfide bond and is able to undergo folding in the IMS (PubMed:16185709, PubMed:19182799, PubMed:21059946, PubMed:23676665). Reduced CHCHD4/MIA40 is then reoxidized by GFER/ERV1 via a disulfide relay system (PubMed:23186364). Mediates formation of disulfide bond in MICU1 in the IMS, promoting formation of the MICU1-MICU2 heterodimer that regulates mitochondrial calcium uptake (PubMed:26387864).

Product Notes
The CHCHD4 chchd4 (Catalog #AAA9603592) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The CHCHD4 Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's CHCHD4 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC). WB: 1:500-1:2000 IHC: 1:50-1:200. Researchers should empirically determine the suitability of the CHCHD4 chchd4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CHCHD4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
