Western Blot (WB)
(Western blot analysis of extracts from BJAB, using CD20 Antibody. The lane on the left was treated with blocking peptide.)
Immunohistochemistry (IHC)-Paraffin
(MBS9612078 at 1/100 staining Human mammary cancer by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary antibody at 4°C overnight. An HRP conjugated anti-Rabbit antibody was used as the secondary antibody.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612078 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612078 1:200) and mouse anti-beta tubulin Ab(T0023 1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Post Translational Modifications: Phosphorylated on serines and threonines in resting B-cells. Protein kinase C/PKC can use CD20 as substrate.
Subcellular Location: Cell membrane>Multi-pass membrane protein. Cell membrane>Lipid-anchor. Note: Constitutively associated with membrane rafts.
Tissue Specificity: Expressed on B-cells.
Subunit Structure: Forms homotetramers (PubMed:7684739, PubMed:18474602). Interacts with the heavy and light chains of cell surface IgM, the antigen-binding components of the BCR (PubMed:18474602).
Similarity: Belongs to the MS4A family.
NCBI and Uniprot Product Information
Predicted Molecular Weight: (Calculated)33kDa.
NCBI Description
This gene encodes a member of the membrane-spanning 4A gene family. Members of this nascent protein family are characterized by common structural features and similar intron/exon splice boundaries and display unique expression patterns among hematopoietic cells and nonlymphoid tissues. This gene encodes a B-lymphocyte surface molecule which plays a role in the development and differentiation of B-cells into plasma cells. This family member is localized to 11q12, among a cluster of family members. Alternative splicing of this gene results in two transcript variants which encode the same protein. Uniprot Description
MS4A1: This protein may be involved in the regulation of B-cell activation and proliferation. Defects in MS4A1 are the cause of immunodeficiency common variable type 5 (CVID5); also called antibody deficiency due to CD20 defect. CVID5 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B-cells is usually in the normal range, but can be low. Belongs to the MS4A family.

Product Notes
The MS4A1 ms4a1 (Catalog #AAA9612078) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The CD20 Antibody reacts with Human Predicted Reactivity: Pig(88%), Bovine(88%), Sheep(88%), Dog(88%) and may cross-react with other species as described in the data sheet. AAA Biotech's CD20 can be used in a range of immunoassay formats including, but not limited to, Immunofluorescence (IF), Immunocytochemistry (ICC), ELISA (EIA). IF: 1:100-1:500 ICC: 1:100-1:500 WB: 1:500-1:2000 IHC: 1:50-1:200 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the MS4A1 ms4a1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD20, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
