Application Notes
Suitable for use in ELISA, Flow Cytometry, Western Blot, Immunohistochemistry and Immunocytochemistry.
Dilution: Flow Cytometry: 1:100
Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4 degree C with gentle shaking, overnight.

Synthetic phosphopeptide corresponding to residues around Ser63 of human c-Jun (KLH coupled).
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

c-Jun is a component of the transcription factor AP-1, which binds and activates transcription at TRE/AP-1 elements. The transcriptional activity of c- Jun is regulated by phosphorylation at Ser63 and Ser73 (1,2). Extracellular signals, including growth factors, transforming oncoproteins and UV irradiation, stimulate phosphorylation of c-Jun at Ser63/73 and activate c- Jun-dependent transcription. Mutation of Ser63/73 renders c-Jun nonresponsive to mitogenic and stressinduced signaling pathways. The MAP kinase homologue SAPK/JNK binds to the N-terminal region of c- Jun and phosphorylates c-Jun at Ser63/73. In addition, the activity of SAPK/JNK is stimulated by the same signals that activate c-Jun (3,4).

Function: Transcription factor that recognizes and binds to the enhancer heptamer motif 5'-TGA[CG]TCA-3'.

Subunit structure: Heterodimer with either FOS or BATF3 or ATF7

By similarity. The ATF7/JUN heterodimer is essential for ATF7 transactivation activity. Interacts with MYBBP1A, SP1, SPIB and TCF20. Interacts with COPS5; indirectly leading to its phosphorylation. Interacts with DSIPI; this interaction inhibits the binding of active AP1 to its target DNA. Interacts with HIVEP3. Component of the SMAD3/SMAD4/JUN/FOS/complex which forms at the AP1 promoter site. The SMAD3/SMAD4 heterodimer acts syngernistically with the JUN/FOS heterodimer to activate transcription in response to TGF-beta. Interacts (via its basic DNA binding and leucine zipper domains) with SMAD3 (via an N-terminal domain); the interaction is required for TGF-beta-mediated transactivation of the SMAD3/SMAD4/JUN/FOS/complex
