Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

BNIP3 (Bcl-2/E1B-19kD interacting prote in 3) is a pro-apoptotic mitochondrial protein and Bcl-2 family member that contains a Bcl-2 homology 3 (BH3) domain a nd a carboxyl terminal transmembrane (TM) domain (1-3). While BNIP3 has a predicted molecular weight of about 22 kD, it runs anomalously on SDS-PAGE and includes a band of around 60 kD that may be a dimeric form that is not re- duced (2). BNIP3 associates with anti-apoptotic family members Bcl-2, Bcl-xL, and the adenovirus homologue E1B-1 9kD. BNIP3 is distinct from other Bcl-2 family members t hat contain only the BH3 domain in that the TM domain, and- not the BH3 domain, is required for mitochondrial targetin- g and pro-apoptotic activity (4). In addition to apoptosis, BNIP3 has been implicated in necrosis (5) and autophagy (6,7), although the relationship between BNIP3 and these processes is not fully understood. Increased expression of BNIP3 under hypoxic conditions is mainly regulated by the transcription factor HIF-1 (8-10). Silencing of the BNIP3 promoter by methylation has been observed in several types of cancer cells and may play an important role in their survival (11-14).

NCBI Description

This gene is a member of the BCL2/adenovirus E1B 19 kd-interacting protein (BNIP) family. It interacts with the E1B 19 kDa protein, which protects cells from virally-induced cell death. The encoded protein also interacts with E1B 19 kDa-like sequences of BCL2, another apoptotic protector. This protein contains a BH3 domain and a transmembrane domain, which have been associated with pro-apoptotic function. The dimeric mitochondrial protein encoded by this gene is known to induce apoptosis, even in the presence of BCL2. [provided by RefSeq]
