Application Notes
BMI-1 antibody can be used for detection of BMI-1 by Western blot at 0.5 to 2 mug/mL. Antibody can also be used for immunocytochemistry starting at 10 mug/mL. For immunofluorescence start at 20 mug/mL.

Preparation and Storage
BMI-1 antibody can be stored at 4 degree C for three months and -20 degree C, stable for up to one year. As with all antibodies care should be taken to avoid repeated freeze thaw cycles. Antibodies should not be exposed to prolonged high temperatures.

BMI-1 Antibody: The transcriptional repressor BMI-1 was first identified as a proto-oncogene frequently activated by Moloney murine leukemia proviral insertions in mice and cooperating with c-myc in the generation of mouse lymphomas. BMI-1 is involved in segment specification, cell growth and maintenance, transcriptional regulation, and chromatin modification. A major target of BMI-1 is the ink4a locus which encodes tumor suppressor proteins p16 and p19Arf, which are important in tumor progression and thought to be critical in cell proliferation and senescence. Recent studies have also shown that BMI-1 is required for the maintenance of adult normal and leukemic stem cells, suggesting that BMI-1 could an attractive therapeutic target for stem cell proliferation and renewal as well as for anti-cancer strategies.

This locus represents naturally occurring read-through transcription between the neighboring COMM domain-containing protein 3 and polycomb complex protein BMI-1 genes on chromosome 10. The read-through transcript produces a fusion protein that shares sequence identity with each individual gene product. [provided by RefSeq, Feb 2011]

BMI1: Component of a Polycomb group (PcG) multiprotein PRC1- like complex, a complex class required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A 'Lys-119', rendering chromatin heritably changed in its expressibility. In the PRC1 complex, it is required to stimulate the E3 ubiquitin-protein ligase activity of RNF2/RING2. Component of a PRC1-like complex. Interacts with RING1 and RING2. Interacts vwith CBX7 and CBX8. Interacts with SPOP. Part of a complex consisting of BMI1, CUL3 and SPOP. Interacts with E4F1.
