This antibody detects endogenous human and mouse BAD only when phosphorylated at serine 112.
Form/Format
100ul at 1.0mg/ml in PBS (without Mg2+ and Ca2+), pH 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol. Affinity-purified on phosphopeptide; non-phosphopeptide- reactive antibodies were removed by chromatography on non-phosphorylated peptide.

Application Notes
Immunoblotting: use at dilution of 1:500- 1:1,000. A ban of ~23kDa is detected.
Immunohistochemistry: use at dilution of 1:50- 1:100.
Dilute in PBS or medium that is identical to that used in the assay system.

The protein encoded by the BAD gene is a member of the Bcl-2 family whose members are regulators of programmed cell death. BAD protein positively regulates cell apoptosis by forming heterodimers with Bcl-xL and Bcl-2 and reversing their death repressor activity. Proapoptotic activity of this protein is regulated through its phosphorylation. Protein kinases AKT and MAP kinase, as well as protein phosphatase calcineurin, were found to be involved in the regulation of this protein. Alternative splicing of this gene results in two transcript variants which encode the same isoform.

Uniprot Description

BAD: a proapoptotic member of the Bcl-2 family. Displaces Bax from binding to Bcl-2 and Bcl-xL, resulting in cell death. Survival factors such as IL-3 can inhibit the apoptotic activity of Bad inducing the phosphorylation of Bad by Akt and p90RSK. 14-3-3 proteins bind phosphorylated Bad, inhibiting its binding to Bcl-2 and Bcl-xL. Phosphorylation by mitochondria-anchored PKA in the BH3 domain can block the dimerization of Bad and Bcl-xL.
