Host
Rabbit
Reactivity
Human
Clonality
Polyclonal
Isotype
IgG
Specificity
This item specifically recognises an epitope within the C-Terminal (CT) region of the Atg5-Atg12 complex, which plays a key role in the regulation of autophagy, and is emerging as an important factor in the innate immune response to viruses.

Studies have shown that the Atg5-Atg12 complex directly interacts with RIG-1 (retinoic acid-inducible gene 1) and the adaptor molecule IPS-1 (IFN-beta promoter stimulator 1), through caspase recruitment domains (CARDs), thereby acting as a negative regulator of the type I IFN production pathway, and as a contributor to RNA viral replication within the host cells.
Form/Format
Purified
Purified IgG - liquid
Concentration
IgG concentration 1.0mg/ml (varies by lot)
Sequence Length
140
Applicable Applications for anti-ATG5-ATG12 antibody
Western Blot (WB)
Application Notes
Western Blot: This item detects a band of approximately 56kDa in mouse liver and human U87 cell lysates.
Western Blotting: Minimum Dilution: 1.0; Maximum Dilution: 2.0ug/ml
Perservative Stabilisers
0.09% Sodium Azide (NaN3)
Immunogen
KLH conjugated synthetic peptide corresponding to amino acids 262-275 of mouse Atg5. The corresponding sequence differs by one amino acid in human.
Buffer Solution
Antiserum Preparation
Antiserum to mouse Atg5 was raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared by affinity chromatography.
Target Species
Mouse
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 18 months from date of despatch.

NCBI Description

Autophagy is a process of bulk protein degradation in which cytoplasmic components, including organelles, are enclosed in double-membrane structures called autophagosomes and delivered to lysosomes or vacuoles for degradation. ATG12 is the human homolog of a yeast protein involved in autophagy (Mizushima et al., 1998 [PubMed 9852036]).[supplied by OMIM, Mar 2008]

Uniprot Description

ATG12: Ubiquitin-like protein required for autophagy. Conjugated to ATG3 and ATG5. Ubiquitous. Belongs to the ATG12 family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Ubiquitin-like modifier; Autophagy

Chromosomal Location of Human Ortholog: 5q21-q22

Cellular Component: pre-autophagosomal structure membrane

Molecular Function: protein binding; APG8 conjugating enzyme activity

Biological Process: mitochondrion degradation; innate immune response; C-terminal protein lipidation; negative regulation of interferon type I production; cellular response to nitrogen starvation; autophagic vacuole formation

Product Notes

The ATG5-ATG12 atg12 (Catalog #AAA224276) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI ATG5-ATG12 COMPLEX (C-TERMINAL) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's ATG5-ATG12 COMPLEX can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Western Blot: This item detects a band of approximately 56kDa in mouse liver and human U87 cell lysates. Western Blotting: Minimum Dilution: 1.0; Maximum Dilution: 2.0ug/ml. Researchers should empirically determine the suitability of the ATG5-ATG12 atg12 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ATG5-ATG12 COMPLEX, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
