Immunohistochemistry (IHC)

(MBS9606789 at 1/100 staining Human prostate tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the antibody for 1.5 hours at 22 degree C. An HRP conjugated goat anti-rabbit antibody was used as the secondary.)

Description: The autophagy-related protein 13 (Atg13) plays an important role in the formation of autophagosomes. Autophagosomes are formed in response to nutrient deprivation and function as the transport vesicles for organelles, proteins, and protein complexes targeted for lysosomes that digest these cargos to produce energy and nutrients. Atg13 is activated by the mTOR pathway and forms a complex with the FIP200 protein. This complex is involved in enhancing the activity of the ULK1 kinase which is required for the formation of autophagosomes. Atg13/FIP200 faciliates the localization of ULK1 to pre-autophagosomes, and subsequently stabilizes ULK1. Autophagy is an important process in development, growth, and cell differentiation, and disruption of this process may contribute to cancer, aging, and neurodegenerative diseases.
Function: Autophagy factor required for autophagosome formation and mitophagy. Target of the TOR kinase signaling pathway that regulates autophagy through the control of the phosphorylation status of ATG13 and ULK1, and the regulation of the ATG13-ULK1-RB1CC1 complex. Through its regulation of ULK1 activity, plays a role in the regulation of the kinase activity of mTORC1 and cell proliferation.

Post-translational Modifications: Phosphorylated by ULK1, ULK2 and mTOR.
