Characterization
To insure acceptable performance, each batch of product is tested by flow cytometry to conform to the characteristics of a standard reference reagent.
Preparation and Storage
Annexin V-UNLB is supplied as 100 tests in 1.0 mL of phosphate buffered saline containing 0.1% PBS/NaN3. Store at 2-8 degree C.
Annexin V-FITC: is supplied as 100 tests in 1.0 mL PBS/NaN3. Store at 2-8 degree C.; protect from light.
Annexin V-BIOT: Store at 2-8 degree C; protect from light.

This s a recombinant protein that is produced in eukaryotic cells.
A number of changes in cell structure occur during apoptosis. One major change is the loss of membrane phospholipid asymmetry, with translocation of phosphatidylserine (PS) from the inner leaflet of the phospholipid bilayer to the external surface.1 Annexin V (Mr 36-kDa), a member of the annexin family of calcium-dependent phospholipid binding proteins, has a high affinity for phosphatidylserine (PS)-containing phospholipid bilayers.2,3. When PS is exposed on the extracellular face of a cell membrane, Annexin V binds with an affinity of approximately 7 nM4. Fluorochrome-conjugated Annexin V can be used to monitor changes in cell membrane phospholipid asymmetry,5-8 thereby providing a convenient tool for detection of apoptotic cells.
Annexin V-FITC is a labeled Annexin V that can be used to evaluate subpopulations of cells along the apoptotic pathway without counterstaining with propidium iodide or other DNA dyes. Analysis is performed on a flow cytometer equipped with a 488 nm argon laser and gated on viable cells by light scatter (see flow histograms below). Alternatively, the cells can be examined by fluorescence microscopy.

NCBI Description

The protein encoded by this gene belongs to the annexin family of calcium-dependent phospholipid binding proteins some of which have been implicated in membrane-related events along exocytotic and endocytotic pathways. Annexin 5 is a phospholipase A2 and protein kinase C inhibitory protein with calcium channel activity and a potential role in cellular signal transduction, inflammation, growth and differentiation. Annexin 5 has also been described as placental anticoagulant protein I, vascular anticoagulant-alpha, endonexin II, lipocortin V, placental protein 4 and anchorin CII. The gene spans 29 kb containing 13 exons, and encodes a single transcript of approximately 1.6 kb and a protein product with a molecular weight of about 35 kDa. [provided by RefSeq]
