Related Product Information for anti-AGRP antibody
Several common diseases such as type II diabetes, hypertension, cardiovascular diseases, hyperlipidemia, and some cancers are associated with obesity. An abnormal increase in body fat relative to lean tissue mass has been used as an indicator of obesity. High fat diet, certain environmental factors, and genetic linkage are the primary causes of obesity. In order to understand the genetic basis of obesity, several monogenic murine obesity models have been characterized including obese (Ob), diabetes (db), fat (fat), agouti yellow (Ay), and tubby (tub). More recently, Tub, the human homolog of mouse Tub, TULP1 & TULP2 (for Tubby Like Proteins) and Agouti related protein (AGRP) have been cloned. The obesity associated with Ay mice may be due to ectopic expression of a secreted protein Agouti. Agouti protein (132 aa in human) is normally expressed in skin but its ubiquitous expression causes obesity. Agouti is a paracrine-signaling molecule that affects pigmentation by inhibiting the melanocortin receptor 1 (MCR-1). However, recombinant Agouti protein also antagonizes the MC2R and MC4R. AGRP (132 aa in human, chromosome 16q21) is normally expressed in adrenal and hypothalamus. AGRP levels are increased several folds in ob/ob mice. AGRP is a strong antagonist of MC3R and MC4R. Ubiquitous expression of AGRP in transgenic mice causes obesity without altering skin pigmentation.

NCBI Description

This gene encodes an antagonist of the melanocortin-3 and melanocortin-4 receptor. It appears to regulate hypothalamic control of feeding behavior via melanocortin receptor and/or intracellular calcium regulation, and thus plays a role in weight homeostasis. Mutations in this gene have been associated with late on-set obesity. [provided by RefSeq]

Uniprot Description

AGRP: Plays a role in weight homeostasis. Involved in the control of feeding behavior through the central melanocortin system. Acts as alpha melanocyte-stimulating hormone antagonist by inhibiting cAMP production mediated by stimulation of melanocortin receptors within the hypothalamus and adrenal gland. Has very low activity with MC5R. Is an inverse agonist for MC3R and MC4R being able to suppress their constitutive activity. It promotes MC3R and MC4R endocytosis in an arrestin-dependent manner. Genetic variations in AGRP may be a cause of obesity (OBESITY). It is a condition characterized by an increase of body weight beyond the limitation of skeletal and physical requirements, as the result of excessive accumulation of body fat.

Product Notes

The AGRP agrp (Catalog #AAA614855) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The AGRP (Agouti Related Protein, ART, AGRT, ASIP2, Agouti-related transcript) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's AGRP can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB). Suitable for use in ELISA and Western Blot. Dilution: ELISA: 1:10,000-1:100,000 using 50-100ng control peptide (A1059-62A) per well. Western blot: 1-10ug/ml using ECL. Researchers should empirically determine the suitability of the AGRP agrp for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "AGRP, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
