Recommended Immunohistochemistry Protocol
The following Protocol is a recommendation only. Results are not guaranteed
Tissue Preperation:
Formalin fixation and embedding in paraffin wax.
Tissue Sectioning:
Make 4-um sections and place precleaned and charged microscope sldies. Heat in a tissue-drying over for 45 Min. at 60°.
Deparaffinization:
Wash dry slides in 3 changes of xylene - 5 min. each side at RT
Rehydration:
Wash slides 3 changes of 100% alcohol - 3 min. each side at RT
Wash slides 2 changes of 95% alcohol - 3 min. each side at RT
Wash slides 1 change of 80% alcohol - 3 min. each side at RT
Rinse slides in gentle running distilled water - 5 min. at RT
Antigen Retrieval:
Steam slides in 0.01 sodium citrate buffer, pH 6.0 at 99-100°C - 20 min
Remove from heat and let stand at RT in buffer - 20 min
Rinse in 1X TBS with tween (TBST) - 1 min. at RT
Immunostaining:
(do not allow tissues to dry at any time during the staining procedure)
Apply a universal protein block - 20 min. at RT
Drain protein block from slides, apply diluted primary antibody - 45 min. at RT
Rinse in 1X TBS with tween (TBST) - 1 min. at RT
Apply biotiylated secondary anitbody appropriate for the primary anitbody - 30 min. at RT
Rinse in 1X TBS with tween (TBST) - 1 min. at RT
Apply alkaline phosphatase streptavidin - 30 min. at RT
Rinse in 1X TBS with tween (TBST) - 1 min. at RT
Apply alkaline phosphatase chromogen substrate - 30 min. at RT
Wash slides in distilled water - 1 min. at RT.
Dehydrate:
(This method should only be used if the chromogen substrate is alcohol insoluble)
Wash slides 2 changes of 80% alcohol - 1 min. each side at RT
Wash slides 2 changes of 95% alcohol - 1 min. each side at RT
Wash slides 3 changes of 100% alcohol - 1 min. each side at RT
Wash slides 3 changes of xylene - 1 min. each side at RT
Apply Covership.
Preparation and Storage
Aliquot and store undiluted at -20°C or below for up to 1 year.
Can be stored undiluted at 4°C for up to 1 month.
Avoid freeze/thaw cycles.

The protein contains a signal sequence, a prodomain with both a potential cysteine switch and a furin cleavage site, a metalloproteinase domain with a conserved zinc-binding motif and a 'met turn,' a disintegrin-like domain, and a spacer region between a thrombospondin type 1 motif and thrombospondin submotif.

NCBI Description

This gene encodes a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) protein family. Members of the family share several distinct protein modules, including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 (TS) motif. Individual members of this family differ in the number of C-terminal TS motifs, and some have unique C-terminal domains. The enzyme encoded by this gene contains two C-terminal TS motifs and functions as aggrecanase to cleave aggrecan, a major proteoglycan of cartilage. [provided by RefSeq, Jul 2008]
