ADAM (a disintegr in and metalloprotease) proteins are a family of over 30 membrane-anchored, glycosylated, Zn2+ dependent proteases that are involved in cell-cell, cell-matrix interface related processes including fertilization, muscle fusion, secretion of TNF (tumor necrosis factor alpha), and modulation of the neurogenic function of Notch and Delta. ADAM proteins possess a signaldomain, a pro-domain, a metalloprotease domain, a disintegrin domain (integrin ligand), a cysteine-rich region, an epidermal growth factor-like domain, a transmembrane domain and a cytoplasmic tail. ADAMs are expressed in brain, testis, epididymis, ovary, breast, placenta, liver, heart, lung, bone and muscle, and catalyze proteolysis, adhesion, fusion and intracellular signaling. This gene encodes a member of a family of proteins that are structurally related to snake venom disintegrins and have been implicated in a variety of biological processes involving cell-cell and cell-matrix interactions, including fertilization, muscle development, and neurogenesis. Expression of this gene has been used as a maternal serum marker for pre-natal development. Alternative splicing results in multiple transcript variants encoding different isoforms. Shorter isoforms are secreted, while longer isoforms are membrane-bound form. [provided by RefSeq, Jan 2014]

ADAM12: contains a disintegrin and metalloprotease (ADAM) domain 12, which is a member of the ADAM protein family. Involved in skeletal muscle regeneration, specifically at the onset of cell fusion, and the differentiation of macrophage-derived giant cells (MGC) and osteoclasts. 2 alternative splicing isoforms have been described: a shorter secreted form and a longer membrane-bound form. The shorter form stimulates myogenesis.
