Suitable for use in ELISA and Western Blot.
Dilution: Western Blot: 1:1000-1:10,000. Detects ACK1 phosphorylated on Tyrosine residue 284 in COS7 cells transfected with wild type ACK1 but not in COS7 cells transfected with ACK1(Y284F). Lysates from COS7 cells, transfected with either wild type ACK1 or mutant ACK1 (Y284F), were resolved by electrophoresis, transferred to PVDF (Immobilon-P), and probed with A0650-01 for 2 hours at RT. Proteins were visualized via HRP and chemiluminescent detection.
Immunogen
Synthetic peptide corresponding to amino acids surrounding Tyr284 of human ACK1 conjugated to KLH.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Ack1 and Ack2 (activated cdc42-associated kinase 1 and 2) are non-receptor tyrosine kinases that consist of a tyrosine kinase core, an SH3 domain, a cdc42/Rac-binding (CRIB) domain, a Ralt homology region and a proline-rich region. Ack1 and 2 are the only two tyrosine kinases known to interact with cdc42. Both Acks are activated by growth factors including EGF and PDGF, as well as by activated integrins through cell adhesion, and may serve to link receptor tyrosine kinase or G protein-coupled receptor signaling with cdc42. Acks may regulate cell growth, morphology and motility. Recent findings indicate that Ack1 may play a role in prostate tumorigenesis, making it a potential drug target for this type of cancer.

NCBI Description

This gene encodes a tyrosine kinase that binds Cdc42Hs in its GTP-bound form and inhibits both the intrinsic and GTPase-activating protein (GAP)-stimulated GTPase activity of Cdc42Hs. This binding is mediated by a unique sequence of 47 amino acids C-terminal to an SH3 domain. The protein may be involved in a regulatory mechanism that sustains the GTP-bound active form of Cdc42Hs and which is directly linked to a tyrosine phosphorylation signal transduction pathway. Several alternatively spliced transcript variants have been identified from this gene, but the full-length nature of only two transcript variants has been determined. [provided by RefSeq]
