Rat phosphorylated microtubule Associated protein tau ELISA Kit
Principle of the Assay: pTAU ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-pTAU antibody and an pTAU-HRP conjugate. The assay sample and buffer are incubated together with pTAU-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the pTAU concentration since pTAU from samples and pTAU-HRP conjugate compete for the anti-pTAU antibody binding site. Since the number of sites is limited, as more sites are occupied by pTAU from the sample, fewer sites are left to bind pTAU-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The pTAU concentration in each sample is interpolated from this standard curve.

Tau: a microtubule-associated protein that regulates microtubule assembly and stability. Apparently involved in the establishment and maintenance of neuronal polarity. Mutations can result in several neurodegenerative disorders such as Alzheimer's disease, Pick's disease, frontotemporal dementia, cortico-basal degeneration and progressive supranuclear palsy. The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker. Axonal polarity is predetermined by tau localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. Nine differentially spliced isoforms have been described. The short isoforms allow plasticity of the cytoskeleton, whereas the longer isoforms may preferentially play a role in its stabilization.
