Intended Uses: This PmaTAU ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human PmaTAU.

Principle of the Assay: PmaTAU ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PmaTAU antibody and an PmaTAU-HRP conjugate. The assay sample and buffer are incubated together with PmaTAU-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PmaTAU concentration since PmaTAU from samples and PmaTAU-HRP conjugate compete for the anti-PmaTAU antibody binding site. Since the number of sites is limited, as more sites are occupied by PmaTAU from the sample, fewer sites are left to bind PmaTAU-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PmaTAU concentration in each sample is interpolated from this standard curve.

NCBI Description

a microtubule-associated protein; expression is found specifically in neurons [RGD, Feb 2006]

Uniprot Description

Tau: a microtubule-associated protein that regulates microtubule assembly and stability. Apparently involved in the establishment and maintenance of neuronal polarity. Mutations can result in several neurodegenerative disorders such as Alzheimer's disease, Pick's disease, frontotemporal dementia, cortico-basal degeneration and progressive supranuclear palsy. The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker. Axonal polarity is predetermined by tau localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. Nine differentially spliced isoforms have been described. The short isoforms allow plasticity of the cytoskeleton, whereas the longer isoforms may preferentially play a role in its stabilization.

Product Notes

The Human pMAPT/pTAU mapt (Catalog #AAA722317) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA722317 ELISA Kit recognizes Human pMAPT/pTAU. It is sometimes possible for the material contained within the vial of "phosphorylated microtubule Associated protein tau, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
