Principle of the Assay||This ELISA kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human PLEKHO1. During the reaction,Human PLEKHO1 in the sample or standard competes with a fixed amount of biotin-labeled Human PLEKHO1 for sites on a pre-coated Monoclonal antibody specific to Human PLEKHO1. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of Human PLEKHO1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Uniprot Description

PLEKHO1: Plays a role in the regulation of the actin cytoskeleton through its interactions with actin capping protein (CP). May function to target CK2 to the plasma membrane thereby serving as an adapter to facilitate the phosphorylation of CP by protein kinase 2 (CK2). Appears to target ATM to the plasma membrane. Appears to also inhibit tumor cell growth by inhibiting AKT- mediated cell-survival. Also implicated in PI3K-regulated muscle differentiation, the regulation of AP-1 activity (plasma membrane bound AP-1 regulator that translocates to the nucleus) and the promotion of apoptosis induced by tumor necrosis factor TNF. When bound to PKB, it inhibits it probably by decreasing PKB level of phosphorylation. 2 isoforms of the human protein are produced by alternative splicing.
