Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate PERLD1 in samples. An antibody specific for PERLD1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPERLD1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PERLD1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PERLD1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene encodes a glycosylphosphatidylinositol (GPI)-specific phospholipase that primarily localizes to the Golgi apparatus. This ubiquitously expressed gene is predicted to encode a seven-transmembrane protein that removes unsaturated fatty acids from the sn-2 position of GPI. The remodeling of the constituent fatty acids on GPI is thought to be important for the proper association between GPI-anchored proteins and lipid rafts. The tethering of proteins to plasma membranes via posttranslational GPI-anchoring is thought to play a role in protein sorting and trafficking. Mutations in this gene cause an autosomal recessive form of neurologic hyperphosphatasia with cognitive disability (HPMRS4).
