Specificity
The peptide is specifically recognized by anti-Sema3A (N-terminal) antibody (MBS472059) in ELISA, and has been shown to block the reactivity of (MBS472059) during Western blot. In addition, the peptide is recommended for use in blocking (MBS472059) reactivity in immunocytochemistry.
Form/Format
Blocking Peptide is supplied in 50ul phosphate-buffered saline and 0.05% sodium azide.
Sequence Length
771
Applicable Applications for SEMA3A peptide
ELISA, Blocking
Application Notes
Blocking: 1:1,000
ELISA: 50 ng/well

End user should determine optimal concentration dependent on the concentration of the antibody.
Recommended for blocking antibody reactivity in Western Blot and Immunocytochemistry, ELISA established in 96-well Nunc Immunoplates where peptide was bound to plates for 2 hours in 0.1 M sodium carbonate buffer, pH 8.5.
Peptide Sequence
Semaphorin 3A synthetic peptide corresponds to amino acids in the Nterminal region of human Sema3A. This sequence is identical to similar regions in rat and mouse Sema3A, and has low homology to other semaphorin family members.
Preparation and Storage
Store at -20°C. Stable for 1 year.
Related Product Information for SEMA3A peptide
One family of inhibitory axon guidance molecules is the semaphorins. The semaphorins include secreted, transmembrane, and GPIanchored extracellular molecules that are involved in regulating axon guidance by inhibiting axons from growing toward incorrect targets. Semaphorin 3A (Sema3A) may play a particularly interesting role in limiting axon regeneration since it is expressed in meningeal fibroblasts that invade the injured spinal cord and surround the glial scar. In addition, the Sema3A co-receptors, Neuropilin -1 and Plexin-A1, are expressed on axons that regenerate up to the injured region, but do not cross this Sema3A-containing region. Thus, Sema3A and its co-receptors may have important roles in regulating axon guidance during neuronal development and after neuronal injury.

NCBI Description

This gene is a member of the semaphorin family and encodes a protein with an Ig-like C2-type (immunoglobulin-like) domain, a PSI domain and a Sema domain. This secreted protein can function as either a chemorepulsive agent, inhibiting axonal outgrowth, or as a chemoattractive agent, stimulating the growth of apical dendrites. In both cases, the protein is vital for normal neuronal pattern development. Increased expression of this protein is associated with schizophrenia and is seen in a variety of human tumor cell lines. Also, aberrant release of this protein is associated with the progression of Alzheimer's disease. [provided by RefSeq]

Uniprot Description

Function: Induces the collapse and paralysis of neuronal growth cones. Could serve as a ligand that guides specific growth cones by a motility-inhibiting mechanism. Binds to the complex neuropilin-1/plexin-1

Subcellular location: Secreted

Domain: Strong binding to neuropilin is mediated by the carboxy third of the protein.

Sequence similarities: Belongs to the semaphorin family.Contains 1 Ig-like C2-type (immunoglobulin-like) domain.Contains 1 PSI domain.Contains 1 Sema domain.

Product Notes

The SEMA3A sema3a (Catalog #AAA474063) is a Peptide and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's Semaphorin-3A (N-terminal) Peptide can be used in a range of immunoassay formats including, but not limited to, ELISA, Blocking. Blocking: 1:1,000 ELISA: 50 ng/well End user should determine optimal concentration dependent on the concentration of the antibody. Recommended for blocking antibody reactivity in Western Blot and Immunocytochemistry, ELISA established in 96-well Nunc Immunoplates where peptide was bound to plates for 2 hours in 0.1 M sodium carbonate buffer, pH 8.5. Researchers should empirically determine the suitability of the SEMA3A sema3a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Semaphorin-3A (N-terminal) Peptide, Peptide" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
