Principle of the Assay: PDGFA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PDGFA antibody and an PDGFA-HRP conjugate. The assay sample and buffer are incubated together with PDGFA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PDGFA concentration since PDGFA from samples and PDGFA-HRP conjugate compete for the anti-PDGFA antibody binding site. Since the number of sites is limited, as more sites are occupied by PDGFA from the sample, fewer sites are left to bind PDGFA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PDGFA concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the platelet-derived growth factor family. The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a motif of eight cysteines. This gene product can exist either as a homodimer or as a heterodimer with the platelet-derived growth factor beta polypeptide, where the dimers are connected by disulfide bonds. Studies using knockout mice have shown cellular defects in oligodendrocytes, alveolar smooth muscle cells, and Leydig cells in the testis; knockout mice die either as embryos or shortly after birth.
