Principle of the assay: Anti-PAP ELISA kit applies the competitive enzyme immunoassay technique utilizing PAP antigen and an Anti-PAP-HRP conjugate. The assay sample and buffer are incubated together with Anti-PAP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Anti-PAP concentration since Anti-PAP from samples and Anti-PAP-HRP conjugate compete for the PAP antigen binding site. Since the number of sites is limited, as more sites are occupied by Anti-PAP from the sample, fewer sites are left to bind Anti-PAP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Anti-PAP concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes an enzyme that catalyzes the conversion of orthophosphoric monoester to alcohol and orthophosphate. It is synthesized under androgen regulation and is secreted by the epithelial cells of the prostate gland. An alternatively spliced transcript variant encoding a longer isoform has been found for this gene. This isoform contains a transmembrane domain and is localized in the plasma membrane-endosomal-lysosomal pathway. [provided by RefSeq, Sep 2008]

Belongs to the histidine acid phosphatase family. 2 isoforms of the human protein are produced by alternative splicing.
