Principle of the Assay: NUP85 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-NUP85 antibody and an NUP85-HRP conjugate. The assay sample and buffer are incubated together with NUP85-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NUP85 concentration since NUP85 from samples and NUP85-HRP conjugate compete for the anti-NUP85 antibody binding site. Since the number of sites is limited, as more sites are occupied by NUP85 from the sample, fewer sites are left to bind NUP85-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NUP85 concentration in each sample is interpolated from this standard curve.

Bidirectional transport of macromolecules between the cytoplasm and nucleus occurs through nuclear pore complexes (NPCs) embedded in the nuclear envelope. NPCs are composed of subcomplexes, and NUP85 is part of one such subcomplex, Nup107-160. [provided by RefSeq, Jul 2008]

NUP85: Essential component of the nuclear pore complex (NPC) that seems to be required for NPC assembly and maintenance. As part of the NPC Nup107-160 subcomplex plays a role in RNA export and in tethering NUP98/Nup98 and NUP153 to the nucleus. The Nup107-160 complex seems to be required for spindle assembly during mitosis. NUP85 is required for membrane clustering of CCL2- activated CCR2. Seems to be involved in CCR2-mediated chemotaxis of monocytes and may link activated CCR2 to the phosphatidyl- inositol 3-kinase-Rac-lammellipodium protrusion cascade. Belongs to the nucleoporin Nup85 family.
