Principle of the Assay: NOS ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-NOS antibody and an NOS-HRP conjugate. The assay sample and buffer are incubated together with NOS-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NOS concentration since NOS from samples and NOS-HRP conjugate compete for the anti-NOS antibody binding site. Since the number of sites is limited, as more sites are occupied by NOS from the sample, fewer sites are left to bind NOS-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NOS concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
Nitric oxide is a reactive free radical which acts as a biologic mediator in several processes, including neurotransmission and antimicrobial and antitumoral activities. Nitric oxide is synthesized from L-arginine by nitric oxide synthases. Variations in this gene are associated with susceptibility to coronary spasm. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May 2009]
Uniprot Description
eNOS: endothelial constitutive nitric oxide synthase. Synthesizes nitric oxide (NO) from arginine and oxygen, which is implicated in vascular smooth muscle relaxation through a cGMP-mediated signal transduction pathway. NO mediates vascular endothelial growth factor (VEGF)-induced angiogenesis in coronary vessels and promotes blood clotting through the activation of platelets. Induced in humans by caloric restriction. Stimulated by calcium/calmodulin. Inhibited by NOSIP and NOSTRIN.

Product Notes
The Bovine NOS nos3 (Catalog #AAA753159) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA753159 ELISA Kit recognizes Bovine NOS. It is sometimes possible for the material contained within the vial of "Nitric oxide synthase, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
