Specificity
The p47 phox (Phospho-Ser370) Phospho Sandwich ELISA Kit is a convenient, lysate- free, high throughput and sensitive assay kit that can detect p47 phox protein phosphorylation and expression profile in cell lysates. The kit can be used for measuring the relative amounts of phosphorylated p47 phox in cultured cells as well as screening for the effects that various treatments, inhibitors (ie. siRNA or chemicals), or activators have on p47 phox phosphorylation.

Principle of the Assay: The Phospho Sandwich p47 phox (Phospho-Ser370) ELISA Kit contains the components necessary for semi-quantitative determination of p47 phox (Phospho-Ser370) concentrations within experimental cell lysate samples. This particular immunoassay utilizes the traditional "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope p47 phox (Phospho-Ser370) while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a colorimetric reaction to ensue upon substrate addition. When the substrate TMB (3, 3', 5, 5'-Tetramethylbenzidine) is added, the reaction catalyzed by peroxidase yields a blue color that is representative of the antigen concentration. Upon sufficient color development, the reaction can be terminated through addition of Stop Solution (2 N Sulfuric Acid) where the color of the solution will turn yellow. The absorbance of each well can then be read by a spectrophotometer where data analysis may begin.

NCBI Description

The protein encoded by this gene is a 47 kDa cytosolic subunit of neutrophil NADPH oxidase. This oxidase is a multicomponent enzyme that is activated to produce superoxide anion. Mutations in this gene have been associated with chronic granulomatous disease. [provided by RefSeq, Jul 2008]

Uniprot Description

p47phox: the 47-kilodalton cytosolic subunit of the multi-protein complex known as NADPH oxidase found in neutrophils. The holo-oxidase produces a burst of superoxide which is delivered to the lumen of the neutrophil phagosome. Contains 2 SH2 domains. Mutations in NCF1, as well as in other NADPH oxidase subunits, can result in chronic granulomatous disease.

Product Notes

The Human, Mouse, Rat NCF1 ncf1 (Catalog #AAA9511040) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9511040 ELISA Kit recognizes Human, Mouse, Rat NCF1. It is sometimes possible for the material contained within the vial of "p47 phox, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
