Principle of the Assay: The Mouse CD110 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse CD110 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Mouse CD110 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD110 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CD110 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD110 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: The thrombopoietin receptor (TPOR), also known as CD110, is a protein that in humans is encoded by the MPL (myeloproliferative leukemia virus) oncogene. The human MPL gene is mapped to chromosome 1p34. The protein encoded by the c-mpl gene, CD110, is a 635 amino acid transmembrane domain, with two extracellular cytokine receptor domains and two intracellular cytokine receptor box motifs. TPO-R deficient mice were severely thrombocytopenic, emphasizing the important role of CD110 and thrombopoietin in megakaryocyte and platelet formation. Upon binding of thrombopoietin CD110 is dimerized and the JAK family of non-receptor tyrosine kinases, as well as the STAT family, the MAPK family, the adaptor protein Shc and the receptors themselves become tyrosine phosphorylated.

Uniprot Description

MPL: a hematopoietic receptor for thrombopoietin. Thrombopoietin is the major regulator of megakaryocytopoiesis and platelet formation. Dimerizes upon binding of thrombopoietin, leading to its phosphorylation and activation of JAKs and STATs. May represent a regulatory molecule specific for TPO-R-dependent immune responses. Two splice-variant isoforms have been described.
