Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for TPOR elisa kit
Description: This Mouse Thrombopoietin receptor (MPL) ELISA Kit employs a two-site sandwich ELISA to quantitate MPL in samples. An antibody specific for MPL has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MPL present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MPL is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MPL bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: In 1990 an oncogene, v-mpl, was identified from the murine myeloproliferative leukemia virus that was capable of immortalizing bone marrow hematopoietic cells from different lineages. In 1992 the human homologue, named, c-mpl, was cloned. Sequence data revealed that c-mpl encoded a protein that was homologous with members of the hematopoietic receptor superfamily. Presence of anti-sense oligodeoxynucleotides of c-mpl inhibited megakaryocyte colony formation. The ligand for c-mpl, thrombopoietin, was cloned in 1994. Thrombopoietin was shown to be the major regulator of megakaryocytopoiesis and platelet formation. The protein encoded by the c-mpl gene, CD110, is a 635 amino acid transmembrane domain, with two extracellular cytokine receptor domains and two intracellular cytokine receptor box motifs. TPO-R deficient mice were severely thrombocytopenic, emphasizing the important role of CD110 and thrombopoietin in megakaryocyte and platelet formation. Upon binding of thrombopoietin CD110 is dimerized and the JAK family of non-receptor tyrosine kinases, as well as the STAT family, the MAPK family, the adaptor protein Shc and the receptors themselves become tyrosine phosphorylated.

Uniprot Description

MPL: a hematopoietic receptor for thrombopoietin. Thrombopoietin is the major regulator of megakaryocytopoiesis and platelet formation. Dimerizes upon binding of thrombopoietin, leading to its phosphorylation and activation of JAKs and STATs. May represent a regulatory molecule specific for TPO-R-dependent immune responses. Two splice-variant isoforms have been described.

Protein type: Membrane protein, integral; Receptor, cytokine

Cellular Component: cell soma; cell surface; Golgi apparatus; neuron projection; plasma membrane

Biological Process: homeostasis of number of cells; negative regulation of apoptosis; platelet formation; positive regulation of lymphocyte proliferation; regulation of chemokine production

Product Notes

The Mouse TPOR mpl (Catalog #AAA9717584) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9717584 ELISA Kit recognizes Mouse TPOR. It is sometimes possible for the material contained within the vial of "Thrombopoietin Receptor (MPL), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
