Intended Uses: This YOD1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse YOD1. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: YOD1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-YOD1 antibody and an YOD1-HRP conjugate. The assay sample and buffer are incubated together with YOD1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the YOD1 concentration since YOD1 from samples and YOD1-HRP conjugate compete for the anti-YOD1 antibody binding site. Since the number of sites is limited, as more sites are occupied by YOD1 from the sample, fewer sites are left to bind YOD1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The YOD1 concentration in each sample is interpolated from this standard curve.

NCBI Description

Protein ubiquitination controls many intracellular processes, including cell cycle progression, transcriptional activation, and signal transduction. This dynamic process, involving ubiquitin conjugating enzymes and deubiquitinating enzymes, adds and removes ubiquitin. Deubiquitinating enzymes are cysteine proteases that specifically cleave ubiquitin from ubiquitin-conjugated protein substrates. The protein encoded by this gene belongs to a DUB subfamily characterized by an ovarian tumor (OTU) domain. Alternative splicing results in multiple transcript variants. YOD1: Hydrolase that can remove conjugated ubiquitin from proteins and participates in endoplasmic reticulum-associated degradation (ERAD) for misfolded lumenal proteins. May act by triming the ubiquitin chain on the associated substrate to facilitate their threading through the VCP/p97 pore. Ubiquitin moieties on substrates may present a steric impediment to the threading process when the substrate is transferred to the VCP pore and threaded through VCP's axial channel. Mediates deubiquitination of both 'Lys-48'- and 'Lys-63'-linked polyubiquitin chains. Able to cleave both polyubiquitin and di- ubiquitin. 2 isoforms of the human protein are produced by alternative splicing.
