Principle of the Assay||The Mouse TIMP4 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse TIMP4 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse TIMP4 coated on a 96-well plate. Standards and samples are pipetted into the wells and TIMP4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse TIMP4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TIMP4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Metalloproteinase inhibitor 4 is an enzyme that in humans is encoded by the TIMP4 gene. This gene belongs to the tissue inhibitor of metalloproteinases gene family. The proteins encoded by this gene family are inhibitors of the matrix metalloproteinases, a group of peptidases involved in degradation of the extracellular matrix. TIMP4 gene is mapped to chromosome 3p25. TIMP4 gene contains 5 exons that span 6 kb of genomic DNA. The gene is expressed as a 1.4-kb transcript abundant in heart and present at low levels in several other tissues. Expressed TIMP4 inhibits MMPs in vitro. What's more, TIMP-4 binds both progelatinase A and the C domain in a similar manner to that of TIMP-2.

NCBI Description

This gene belongs to the TIMP gene family. The proteins encoded by this gene family are inhibitors of the matrix metalloproteinases, a group of peptidases involved in degradation of the extracellular matrix. The secreted, netrin domain-containing protein encoded by this gene is involved in regulation of platelet aggregation and recruitment and may play role in hormonal regulation and endometrial tissue remodeling. Uniprot Description

TIMP4: Complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them by binding to their catalytic zinc cofactor. Known to act on MMP-1, MMP-2, MMP-3, MMP-7 and MMP- 9. Belongs to the protease inhibitor I35 (TIMP) family.

Product Notes

The Mouse TIMP4 timp4 (Catalog #AAA8244561) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8244561 ELISA Kit recognizes Mouse TIMP4. AAA Biotech's TIMP4 can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (EIA). Researchers should empirically determine the suitability of the TIMP4 timp4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TIMP4, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
