Principle of the Assay: The Picokine™ Mouse Sdc3 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Mouse Sdc3 with a 96-well strip plate that is pre-coated with antibody specific for Sdc3. The detection antibody is a biotinylated antibody specific for Sdc3. The kit contains recombinant Mouse Sdc3 with immunogen: Expression system for standard: NSO; Immunogen sequence: A45-V387. The kit is analytically validated with ready to use reagents. To measure Mouse Sdc3, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Mouse Sdc3 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Sdc3 in the sample.
Background/Introduction: Syndecan-3 is a protein that in humans is encoded by the SDC3 gene. The protein encoded by this gene belongs to the syndecan proteoglycan family, which was assigned to human chromosome 1p35.2, and it showed that in the mouse the Synd3 gene maps to chromosome 4 near the Lmyc gene. The transfection of SDC3 resulted in the formation of long filopodia-like structures, microspikes, and varicosities in several cell lines. Besides, the gene plays a role in the organization of cell shape by affecting the actin cytoskeleton, possibly by transferring signals from the cell surface in a sugar-dependent mechanism. Allelic variants of this gene have been associated with obesity.
