Principle of the assay: Sp100 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-Sp100 antibody and an Sp100-HRP conjugate. The assay sample and buffer are incubated together with Sp100-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Sp100 concentration since Sp100 from samples and Sp100-HRP conjugate compete for the anti-Sp100 antibody binding site. Since the number of sites is limited, as more sites are occupied by Sp100 from the sample, fewer sites are left to bind Sp100-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Sp100 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a subnuclear organelle and major component of the PML (promyelocytic leukemia)-SP100 nuclear bodies. PML and SP100 are covalently modified by the SUMO-1 modifier, which is considered crucial to nuclear body interactions. The encoded protein binds heterochromatin proteins and is thought to play a role in tumorigenesis, immunity, and gene regulation. Alternatively spliced variants have been identified for this gene; one of which encodes a high-mobility group protein. [provided by RefSeq, Aug 2011]
