Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for S-100 elisa kit
Description: This Mouse Soluble protein-100 (S-100) ELISA Kit employs a two-site sandwich ELISA to quantitate S100A1 in samples. An antibody specific for S100A1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any S100A1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for S100A1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of S100A1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: A1is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. This protein may function in stimulation of Ca2+-induced Ca2+ release, inhibition of microtubule assembly, and inhibition of protein kinase C-mediated phosphorylation. Reduced expression of this protein has been implicated in cardiomyopathies. S100A1 overexpression enhances cardiac contractile performance which suggests that S100A1 is a regulator of myocardial contractility. S100A1 improves cardiac performance both by regulating calcium ion handling by the sarcoplasmic reticulum and the responsiveness of myofibrils to calcium ion.

Uniprot Description

S100A1: Weakly binds calcium but binds zinc very tightly- distinct binding sites with different affinities exist for both ions on each monomer. Physiological concentrations of potassium ion antagonize the binding of both divalent cations, especially affecting high-affinity calcium-binding sites. Dimer of either two alpha chains, or two beta chains, or one alpha and one beta chain. Interacts with AGER and CAPZA1. Interacts with FKBP4. Interacts with RYR1 and RYR2. Highly prevalent in heart. Also found in lesser quantities in skeletal muscle and brain. Belongs to the S-100 family.

Protein type: Calcium-binding; Endoplasmic reticulum

Cellular Component: I band; neuron projection; sarcoplasmic reticulum; cytoplasm; M band; nucleus; Z disc; A band

Molecular Function: identical protein binding; protein binding; protein homodimerization activity; metal ion binding; calcium ion binding; calcium-dependent protein binding; ATPase binding

Biological Process: negative regulation of transcription from RNA polymerase II promoter; regulation of heart contraction

Product Notes

The Mouse S-100 s100a1 (Catalog #AAA9717101) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9717101 ELISA Kit recognizes Mouse S-100. It is sometimes possible for the material contained within the vial of "Soluble Protein-100 (S-100), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
