Mouse Neurofilament light polypeptide (NEFL) ELISA Kit
Principle of the Assay: NEFL ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-NEFL antibody and an NEFL-HRP conjugate. The assay sample and buffer are incubated together with NEFL-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NEFL concentration since NEFL from samples and NEFL-HRP conjugate compete for the anti-NEFL antibody binding site. Since the number of sites is limited, as more sites are occupied by NEFL from the sample, fewer sites are left to bind NEFL-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NEFL concentration in each sample is interpolated from this standard curve.

Neurofilaments are type IV intermediate filament heteropolymers composed of light, medium, and heavy chains. Neurofilaments comprise the axoskeleton and they functionally maintain the neuronal caliber. They may also play a role in intracellular transport to axons and dendrites. This gene encodes the light chain neurofilament protein. Mutations in this gene cause Charcot-Marie-Tooth disease types 1F (CMT1F) and 2E (CMT2E), disorders of the peripheral nervous system that are characterized by distinct neuropathies.
