Principle of the Assay: Mouse MMP12 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse MMP12 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse MMP12 coated on a 96-well plate. Standards and samples are pipetted into the wells and MMP12 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse MMP12 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MMP12 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Matrix metalloproteinase-12 (MMP12), also known as MME or ME, is an enzyme that in humans is encoded by the MMP12 gene. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.2. It is thought that the protein encoded by this gene is cleaved at both ends to yield the active enzyme, but this processing has not been fully described. The enzyme degrades soluble and insoluble elastin. It may play a role in aneurysm formation and studies in mice suggest a role in the development of emphysema. This gene may involved in tissue injury and remodeling.

Uniprot Description

MMP12: May be involved in tissue injury and remodeling. Has significant elastolytic activity. Can accept large and small amino acids at the P1' site, but has a preference for leucine. Aromatic or hydrophobic residues are preferred at the P1 site, with small hydrophobic residues (preferably alanine) occupying P3. Belongs to the peptidase M10A family.

Product Notes

The Mouse MMP12 mmp12 (Catalog #AAA824765) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA824765 ELISA Kit recognizes Mouse MMP12. AAA Biotech's MMP12 can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (SE). Researchers should empirically determine the suitability of the MMP12 mmp12 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MMP12, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
