Principle of the assay: MTNR1A ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MTNR1A antibody and an MTNR1A-HRP conjugate. The assay sample and buffer are incubated together with MTNR1A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MTNR1A concentration since MTNR1A from samples and MTNR1A-HRP conjugate compete for the anti-MTNR1A antibody binding site. Since the number of sites is limited, as more sites are occupied by MTNR1A from the sample, fewer sites are left to bind MTNR1A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MTNR1A concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes one of two high affinity forms of a receptor for melatonin, the primary hormone secreted by the pineal gland. This receptor is a G-protein coupled, 7-transmembrane receptor that is responsible for melatonin effects on mammalian circadian rhythm and reproductive alterations affected by day length. The receptor is an integral membrane protein that is readily detectable and localized to two specific regions of the brain. The hypothalamic suprachiasmatic nucleus appears to be involved in circadian rhythm while the hypophysial pars tuberalis may be responsible for the reproductive effects of melatonin.
