Principle of the Assay: Mouse Induced myeloid leukemia cell differentiation protein Mcl-1 (MCL1) ELISA Kit employs a two-site sandwich ELISA to quantitate MCL1 in samples. An antibody specific for MCL1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MCL1 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Mouse MCL1 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of MCL1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background/Introduction: Myeloid cell leukemia sequence 1 (MCL1) is involved in the regulation of apoptosis versus cell survival, and in the maintenance of viability but not of proliferation. MCL1 mediates its effects by interactions with a number of other regulators of apoptosis. Two transcripts from the MCL1 gene have been identified. Isoform 1 enhances cell survival, while isoforms 2 promotes apoptosis.
