Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- Galectin-1 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-GALECTIN-1 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Galectin-1 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Galectin-1 can be calculated.

Background: Galectin-1 is a member of the galectins, which are a family of beta-galactoside-binding proteins implicated in modulating cell-cell and cell-matrix interactions. In Mouses it is encoded by the LGALS1 gene, which maps to 22q12-q13. It is expressed by the endometrial stromal cells throughout the menstrual cycle, however significantly increases during implantation. The Galectin-a plays a role in the immunosuppression required for a successful pregnancy, and it may also act as an autocrine negative growth factor that regulates cell proliferation.

Subcellular location: Secreted › extracellular space › extracellular matrix Ref.24.

Tissue specificity: Expressed in placenta, maternal decidua and fetal membranes. Within placenta, expressed in trophoblasts, stromal cells, villous endothelium, syncytiotrophoblast apical membrane and villous stroma. Within fetal membranes, expressed in amnion, chorioamniotic mesenchyma and chorion (at protein level). Expressed in cardiac, smooth, and skeletal muscle, neurons, thymus, kidney and hematopoietic cells. Ref.4
